How styled characters count against the limit depends on how the platform counts. Most styled letters live in a part of Unicode that some systems count as one character and others count as two, which is the same reason an emoji sometimes costs double. Compose the full text, paste it, and check what the field reports before you rely on the count. Fullwidth is the exception to watch: it still costs one character per letter, but each letter takes roughly double the width on screen, so a fullwidth line wraps far earlier than its character count suggests and can quietly consume two lines of your layout.

The more useful constraint is visual rather than technical. A bio rendered entirely in decorative characters is harder to read at the size Instagram displays it, and the effect that makes one styled line stand out disappears when every line is styled. The common approach is to style the first line or the last, and leave the middle plain. The contrast is what does the work.

Worth remembering that Instagram uses bio text when classifying what an account is about. Decorative characters are not the same strings as their plain equivalents, so a bio written entirely in them gives the platform less to work with. Keeping your main descriptive term in plain characters somewhere costs you nothing visually and leaves that signal intact.
